ABSTRACT

OBJECTIVE@#To investigate whether kirenol, the major pharmacologically active compound of the Chinese medicinal herb , can protect mice from dextran sulfate sodium (DSS)-induced ulcerative colitis (UC).@*METHODS@#C57BL/6 mice with or without kirenol pretreatment were treated with DSS in drinking water for 7 days to induce UC. The symptoms of UC including weight loss, diarrhea and bloody stool were observed daily and graded using the disease activity index (DAI). Colon injury of the mice was assessed by measuring the length of the colon and HE staining of the colon tissue. The levels of inflammatory cytokines produced by the mesenteric lymph nodes (MLNs) lymphocytes were measured using enzyme-linked immunosorbent assay; the apoptosis of the lymphocytes and CD4 T cells was analyzed using flow cytometry.@*RESULTS@#The mice receiving pretreatment with kirenol showed obviously ameliorated symptoms of UC and milder pathological changes in the colon as compared with the control mice. Kirenol treatment significantly down-regulated the secretion of IFN-γ, IL-17A, IL-6 and TNF-α by the MLNs lymphocytes and increased the apoptosis of lymphocytes, especially CD4 T cells in the DSS-treated mice.@*CONCLUSIONS@#Kirenol can protect against T cell-mediated colon injury in DSS-treated mice possibly by suppressing the secretion of inflammatory mediators and inducing apoptosis of the inflammatory lymphocytes.

ABSTRACT

ObjectiveTo determine the Golgi protein-73 (GP73) level and platelet (PLT) count in the peripheral blood of patients with hepatitis B virus (HBV)-related liver diseases and to investigate their values in diagnosing different stages of HBV-related liver diseases. MethodsPeripheral blood samples were collected from 215 patients treated in Beijing Youan Hospital, including 36 patients with chronic hepatitis B (CHB), 32 patients with liver cirrhosis (LC), and 147 patients with hepatocellular carcinoma (HCC). The level of GP73 was measured by ELISA and the PLT count was determined using the Automatic Hematology Analyzer. Comparison of continuous variables was performed using one-way ANOVA, and comparison of categorical variables using chi-square test. The correlation between different variables was assessed by liner regression analysis. ResultsGP73 level (ng/ml) and PLT count (×109/L) displayed similar changes between patients with CHB, LC, pro-early HCC, early HCC, progressive HCC, and advanced HCC. The GP73 level and PLT count were significantly lower in patients with pro-early HCC(80.5±49.8; 84.4±40.0) than in those with CHB(135.5±95.8; 174.4±49.9), progressive HCC(133.8±71.1; 122.0±70.50), and advanced HCC(192.2±98.1; 176.2±86.3) (all P<0.01). ConclusionGP73 level and PLT count change in patients with different stages of HBV-related diseases. The dynamic monitoring of GP73 level and PLT count is useful in the early diagnosis of HCC and the prognostic judgment of HCC patients.

Insulin-like growth factor-I (IGF-I) is a multifunctional cell regulating factor, which plays an important role in the normal physiological activities and pathological process, This article reviewed the research status of the molecular structure of IGF- I and its regulatory factors, the molecular biological role of IGF- I in cell proliferation, cell motility, apoptosis, and reviewed the important role of IGF-I in head and neck tumor, as well as put forward the prospects of IGF-I in middle ear cholesteatoma pathogenesis.

ABSTRACT

Objective To detect the level of visfatin in the peripheral blood ofpafients with coronary atherosclerotic heart disease (coronary heart disease),and investigate the conelation between the level of visfatin and the lesion degree of coronary heart disease.Methods Two hundred and twenty patients who were diagnosed as coronary heart disease by coronary artery angiography from January to June 2011 (coronary heart disease group) were enrolled in this study,including 74 cases with stable angina pectoris(stable angina pectoris group),60 cases with unstable angina pectoris (unstable angina pectoris group),and 86 cases with acute myocardial infarction (myocardial infarction group).And 20 healthy persons with normal coronary artery angiography were selected as control group.The biochemical parameters of triglyceride (TG),total cholesterol (TC),high density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C) were detected by biochemistry autoanalyzer.The level of serum visfatin was detected by ELISA analysis.The serum biochemical parameters and serum visfatin among all the groups were compared and the correlation between them was analyzed.ResuIts The level of serum visfatin of coronary heart disease group [(34.07 ±5.51) μg/L] was significantly higher than that of control group [(13.22 ±3.17) μg/L](P<0.05).TC and LDL-C of different coronary heart disease groups had no significant differences compared with that of control group (P > 0.05).TG,H DL-C and serum visfatin of stable angina pectoris group,unstable angina pectoris group and myocardial infarction group [(1.44 ±0.27) mmol/L,(1.16 ±0.12) mmol/L,(21.36 ± 3.35) μg/L; ( 1.84 ±0.32) mmol/L,(1.01 ± 0.08) mmol/L,(27.78 ±4.47) μg/L; (2.31 ±0.34)mmol/L,(0.93 ± 0.06) mmol/L,(33.14 ± 5.66) μ g/L] had statistical significance compared with those of control group [(0.93 ±0.25) mmol/L,(1.48 ± 0.24) mmol/L,(13.22 ±3.17) μg/L](P<0.05 or <0.01).TG,C,ensini score and serum visfatin of unstable angina pectoris group were significantly higher than those of stable angina pectoris group and HDL-C was obviously lower than that of stable angina pectoris group (P <0.05).Gensini score and serum visfatin of myocardial infarction group were significantly higher than those of unstable angina pectoris group and HDL-C was obviously lower than that of unstable angina pectoris group (P < 0.05).Spearman correlation analysis showed that the level of serum visfatin of coronary heart disease group was positively correlated with TG and Gensini score (P <0.05 or <0.01 ) and negatively correlatedwith HDL-C (P < 0.01 ),and had no correlation with TC and LDL-C (P > 0.05).Conclusions The highlevel expression of visfatin in the peripheral blood may be a risk factor of coronary heart disease.The changes of serum visfatin can reflect the lesion severity degree of coronary artery.

ABSTRACT

<p><b>OBJECTIVE</b>To study the difference in stress and apoptosis related genes transcription between hTERT-RPE1 cells exposed to simulated microwave radiation and the cells with heat water bath, and the effects of microwave on gene transcription in cultured human retina pigment epithelial cells.</p><p><b>METHODS</b>cDNA microarray technique was used to detect the mRNA isolated from hTERT-RPE1 cells exposed to 2 450 MHz simulated microwave radiation and with heat water bath, respectively.</p><p><b>RESULTS</b>Among the 97 related aim genes, there were seven genes up-regulating its transcription, i.e., M31166 (2.52fold), L24123 (2.66fold), AF039704 (2.22fold), U67156 (2.07fold), AF040958 (2.13fold), NM-001423 (2.63fold) and NM-005346 (3.68fold). But, no notably down-regulating gene in transcription was detected.</p><p><b>CONCLUSIONS</b>Microwave could induce up-regulating in multiple stress and apoptosis related genes transcription in cultured human retina pigment epithelial cells, hTERT-RPE1 cells. Microwave radiation has unique effect itself in addition to its heat effect.</p>
